Description |
During progesterone-stimulated Xenopus oocyte maturation, certain pre-existing (maternal) mRNAs are selectively translated in a strict temporal order. The early translational activation of the Mos mRNA requires a Musashi binding element (MBE) that is located in the Mos 3’ untranslated region (UTR). The MBE lies within a 24 nt region originally termed a polyadenylation response element (PRE), but further analyses demonstrated that the MBE within this sequence was critical for temporal regulation (Charlesworth et.al., 2002; Charlesworth et.al., 2006). The MBE consensus sequence (Imai et.al., 2001) is represented in a number of other Xenopus maternal mRNAs as well as in mRNA 3’ UTRs of vertebrate species. The early acting MBE is functionally distinct from the late acting cytoplasmic polyadenylation elements (CPEs). The ability of the MBE to direct early translational activation in Xenopus requires interaction with Musashi, a known stem cell self renewal factor (Charlesworth et.al, 2006; Arumugam et.al., 2010). It should be noted that in mammalian somatic cells Musashi appears to facilitate repression of MBE-containing mRNAs, suggesting a context-dependent regulation of Musashi translational control. |
