Description |
In eukaryotes the formation of the 3'end of mRNA involves cleavage of the nascent transcript and polyadenylation. Two sequence motifs provide the signals that determine the site of polyadenylation. i) the AAUAAA motif typically located 15-20 nt upstream the cleavage site (see as an example the TPM2 mRNA in the Transcript(s)/Gene(s) section below), and ii) an immediately downstream U/GU-rich sequence. These sequence motifs are bound by two protein complexes, the Cleavage and Polyadenylation Stimulation Factor (CPSF) and the Cleavage Stimulation Factor (CstF) (Proudfoot et al., 2002). The CPSF160 subunit has been shown to directly bind the polyadenylation signal (PAS) AAUAAA, which may also occur in one of the following variants: AUUAAA, AGUAAA, UAUAAA, CAUAAA, GAUAAA, AAUAUA, AAUACA, AAUAGA, AAUGAA, ACUAAA, AACAAA, UUUAAA (Beaudoing et al., 2000). Other cis-regulatory signals located upstream and downstream PAS may regulate tissue-specific alternative polyadenylation which has been detected in approximately 54% of human genes (Zhang et al., 2005). |
