Description |
Apolipoprotein B100 (apoB) is an important protein synthesized in liver that mediate uptake and clearance of very low density lipoprotein (VLDL) and low density lipoprotein (LDL) from plasma. Increased levels of VLDL and LDL in plasma correlate with the development of coronary artery diseases and atherosclerosis. The translation efficiency of human apoB mRNA is increased by a cis-regulatory element located in the 5'UTR (Pontrelli et al., 2004). The human apoB 5'UTR is 128 nucleotides long, 76% GC rich, and has several predicted stem loop structures. Recent studies indicated that insulin regulates apoB mRNA translation by decreasing translational efficiency through the apoB 5′UTR. This regulatory activity may involve the binding of a 110-kDa trans-acting protein factor, which is observed to decrease in the presence of insulin (Sidiropoulos et al., 2007). The apoB 5'UTR is remarkably conserved in chimp (XM_001141877), rhesus (XM_001097500), mouse (BC038263) and rat (CB705521) apoB mRNAs, even if apparently there is no significant structural conservation as determined by RNAz application. |
