Description

S-Adenosylmethionine Decarboxylase is a key regulatory enzyme which catalyzes a step in the biosynthesis of the polyamines spermidine and spermine. This step is regulated by a feed-back mechanism dependent on the level of polyamines in the cell. The mRNA of AMD1 contains an uORF encoding a small hexapeptide (MAGDIS) that interacts with the peptidil transferase complex, inhibiting the release of the ribosome from the translational apparatus. Site-directed mutagenesis has been used to demonstrate that the sequence of the uORF peptide is important for the inhibitory regulation of AMD1.

This uORF has been characterized in human, and is also conserved in mouse, cow, cock and zebrafish mRNAs.
